(i) Hexane is a non-polar solvent and ethyl acetate is polar solvent. 90:10 hexane:ethyl acetate means the solvent is more non-polar. When this solvent is used, it eluted beta-carotene. So, beta carotene would be also non-polar. If we use only hexane as solvent, we will get pure beta-carotene. Spots of other compounds in the sample will not be obtained.
(ii) After we elute beta-carotene completely, we will add mixture of hexane: ethyl acetate in ratio of 80:20. More of hexane will be used, because ethyl acetate will start eluting xanthophyll also. To get pure chlorophyll, 80:20 mixture of solvent will be used.
(iii) To elute xanthphyll, pure ethyl acetate will be used.
