To evaluate inhibitory effects of the selected molecules (A1-A15), 10mM stock solutions of each molecule were prepared in DMSO. A reaction mixture (200ul) was prepared with the same formula optimized for the enzyme assay (0.1 M Tris-HCl pH 8, 0.1 M KCl, 25 mM NaCl, 0.25 mM ATP, and two units of inorganic yeast pyrophosphotase) with 10 uM of the sample molecule. The reaction mixture was incubated for 20 minutes at ambient temperature. Absorbance of the product was monitored at 290 nm for 10 minutes after the addition of Substrate B (0.2M).

Six out of 15 sample molecules showed appreciable inhibition at 10 um (Figure 5). Three of the molecules, A3, A6, and A7, exhibited more than 50% inhibition of the enzyme activity and were further diluted to find the minimal inhibitory concentration (MIC). Molecules A3 and A6 exhibited 30% and 45% inhibition of the enzyme activity, respectively, even at 1 um. DMSO was found not to interfere with the enzyme.

Results: Results are given accuarately but the activity ofthe test compounts should be compared with a suitable standard (of known best activity), Results of standard must be included.

Interpretation of results: As A3, A6, and A7, exhibited more than 50% inhibition of the enzymeactivity, they are having good enzyme inhibitory activiy.
