For Microbiology, I am doing a lab and am stuck on two of the questions. It is 1-4 Common Aseptic Transfers and Inoculation Methods.

1) Which medium was most difficult for you to transfer from? Which medium was most difficult for you to inoculate? Explain your difficulties. (Note: there are no current answers to these questions, but based on your evaluation of personal experiences)

2) Did you get growth on/in the sterile NB and NA slant tubes you practiced with? If so, where did you see it and what might have been its source(s)?

1. most difficult to transfer from is agrobacterium and most difficult to transfer is yeast compared to e.coli. because agrobactreium is slowest to grow and cloning is also no very much positive result. more over ligation is problematic in agrobactrium.

2. nutrieent broth and nutirent agar media need maximum sterility conditions because it is very favourable condition for growth of contaminanting organisms. the source could be from yourhand gloves itself or not poperly cleaned hood, or not clean pipette tip box where you store the sterile tips or many other things on the work benck. during the work with NB and NA, it is recommended to do the maximum possible under clean and functional sterile laminar air flow.
