Question
The absorption spectra for chlorophyll a and chlorophyll b are plotted below on the same axis. If these were a mixed sample, how could you determine which is which? If these were mixed how might you only get chlorophyll a and prove that you were correct in only having chlorophyll a?
Explanation / Answer
UV-Vis spectra of chlorophyll
Chlorophyll b absorbs at shorter wavelength as compared to chlorophyll a. Thus among the two peaks, the one appearing before is for chlorophyll b round 645 nm and the one after is chlorophyll a around 660 nm.
To confirm this, one may add a standard sample of chlorophyll a or chlorophyll b, one at a time and see the increase in intensity of the peak with each addition. This will proove which peak corresponds to which chlorophyll.
