Question
The data to the right were collected for the myosin-catalyzed hydrolysis of ATP. Use these data to determine the values of Rmax (the maximum rate of the enzyme, often called the maximum velocity, Vmax) and KM, the Michaelis-Menten constant. Answer the four parts below.
The data to the right were collecte for the myosin-Catalyzed hydrolysis of ATP. Use these data to determine the values of 75 67.0 Rmax (the maximum rate of the enzyme, often called the 125 95.0 maximum velocity, Vmax) and KM, the Michaelis Menten constant. Answer the four parts below 20.0 119 435 155 62.5 166 a) If you were to plot this data to graphically determine Rmax and KM using a Lineweaver-Burk plot, what would you plot for the following data? If you would plot the original data, enter the original value in the corresponding blank ATP graph as graph as pmol L s umol. L Number Number 125 008 95.0 0.01053 L. s. pmol L. pmol Number Number 62.5 0.016 166 0.00602 L. s. pmol L. pmol b) Calculate the slope using the data points from part (a). Then determine the yintercept using the second point. Number Number L. s pmol y-intercept c) Use the y-intercept to calculate RmaxExplanation / Answer
answer b) we use slope formula
slope of graph is equal to = delta Y/ delta X = Y2 -Y1 / X2 - X1
we put values from part (A) and put it in above we get
0.016 - 0.08 / 0.00602 - 0.01053 = 14.19 s
we use y = mx+c (equation of straight line ) to find intercept
0.01053 = 14.19 * 0.08 + C
C= 0.01053 - 1.1352 = -1.124
and for Vmax we know at x=0 , y = 1/ Vmax
so Vmax or Rmax = 1/y = 1/ 1.124 = 0.8896 L.s.mol-1
Lineweaver-Burk Equation
1/V = Km / Vmax or Rmax * 1/S + 1/Vmax
