7. If you had a loss of function mutation in the methylated DNA binding protein MECP2, what pattern of activity (if any) would you expect to see in your imprinted genes? How does this differ (if it does) from the pattern of activity you are supposed to have in these genes?
8. If an interfering RNA interfered with the production of the transformer (Tra) protein in Drosophila (fruitflies), what ratio of male to female fruitflies would you expect to see? Why?

7. MEP2 binds to CpG islands containing methyl groups and is most likely involved in gene silencing. Thus, the imrinted genes which are meant to be silenced in a cell would be derepressed and hence would be expressed. This expression might impair the epigenetic regulation which was supposed to express the genes in a controlled and regulated manner. This impairment may lead to a diseased condition.
8. Transformer protein is a female specific protein transformer that controls female somatic sexual differentiation. Hence the ratio of males will increase as tra protein will be unable to splice the target mRNA in a female specific manner. Please refer Gilbert for a better understanding.
