As you take vitals she explains that they immigrated from Pakistan just last week. Her son had diarrhea for several days but she assumed it was the stress from travel. There are militant groups in Pakistan that kidnap and murder health care workers who administer the polio vaccine. For this reason the boy has not been vaccinated. Design an ELISA that will test whether the boy has polio (2 points). Explain what would happen if the boy were positive for polio virus (2 points) or negative for polio virus (2 points). Hint – polio virus is shed in the stool.
Explanation / Answer
Our body produces IgG antibodies against polio viruses. In an ELISA method, we will coat the plate ith polio antibodies and antigens will be presented to these antibodies, Again these antigens will be bind with an enzyme linked antibody. So, when we add substrate to these enzyme-linked antibodies, they catalyse the reaction. Based on the amount of product prouced, we can determine the presence of specific antibody. This is the sandwitch ELISA method.
