Question

please help me solve this problem thank you A temperate bacteriophage has the gene order xl x2 x3 att x4 x5. What is the gene order of the prophase Can the lytic PI phage infect the lysogen produced by integration of the phage described in question a? Explain your answer. A new series of all mutations in the virulent phage T4 has been isolated, and the results of phage recombination experiments between all twelve of the new mutations are listed below: Draw a linear genetic map showing the location of all point mutations and the frequency of recombination between adjacent mutations. Describe the location and extent of all deletions.

Explanation / Answer

a)

att x 4 x 5 (cos site) x 1 x 2 x 3 x att. Remember, during integration of phage in bacterial chromosome the att sites cross in opposite way at site of recombination.

b)

No, a lytic phage can not infect a lysogen produced by integration of phage because after during a after integration the lysogen produces a repressor protein coded by phage gene. e.g. in bacteriophage lambda there exists a competition between repressor protein and cro protein (inducer of late phage genes) and whosoever wins the race brings out the outcome of lytic / lysogenic cycle. cro in excess can repress the promoter of repressor protein and vice versa. As lysogens already have excess repressor protein, so these cells are immune to lytic phages.
