Question

These Questions are all in regards to Psychological characteristics of bacteria and streaking for isolation.

Part A

1) How is oxygen removed in the GasPak anaerobic jar?

2) What process is more efficient in releasing energy from glucose, anaerobic or aerobic respiration? Explain

3) When yeast is grown as a bulk source of vitamins, it is grown aerobically. When is it grown in wine production, it is kept anaerobic. Why?

Part B

1) What is an advantage of solid culture over liquid culture?

2) Give 2 reasons why the nutrient agar should be cooled to 45 degrees C.

3) What temperature does agar melt?

4) What temperature does agar solidify?

5) Why are plates inverted during incubation?

Explanation / Answer

1. The GasPak is used to create anaerobic environment, and is often used in experiment procedures. In case of growing anaerobic microbes, the inoculated plates are introduced into the chamber. Water is added to gas generator envelop placed inside the jar before sealing the jar.

The gas generator envelop contains sodium borohydride and sodium bicarbonate. Reaction of water with these chemicals release hydrogen and carbon dioxide gases. Hydrogen reacts with the free oxygen present in the jar to produce water, and thus consumes all the oxygen, which creates oxygen free environment. This reaction is catalyzed by palladium, which is attached to the lid of the jar. Bow, the oxygen is replaced by CO2.
