The enzyme is a biological catalyst that binds to the substrate molecules and increase the rate of reaction.

In a recation , where the all the active sites of the enzyme are not yet saturated , increasing the concentration of substrate will increase the rate of reaction. But as the concentration is increased constantly this effect will become little and little and after some time there will be no effect on the reaction velocity even though the substrate concentration is increased many folds.

This is due to a simple reason that earlier active sites of enzymes were empty , so when concentration was increased , substrate occupied the active sites and got converted into products. Slowly and gradually the active sites get saturated , due to which no more substrate is able to bind to the enzyme , hence the reaction rate does not seem to increase at all.
