In strong binding variant, two OH group is attached in a seven membered ring and they have a specific stereochemistry. That implies the strong binding variant has a rigid structure which is exactly fitted with HIV protease inhibitor. Hence enthalpy of binding is less in this case.

Whereas weak binding variant has an open structers for two OH group. So to bind with HIV protease inhibitor it nedd some conformational changes. Hence it requires more energy to bind, i.e., more enthalpy.

Hence strong site is more favourable than weak side.
