Question

theres a showing of an Oc mutant of the lac operon. This stands for a constitutive operator mutant.

a) if you purified DNA fragments of wild type and Ocversions of the operator and purified lac repressor, describe how you might use a nitrocellulose filter binding assay to compare how the repressor would bind the two types of operator.

b) Describe how the results of DNAse footprinting assay might look using repressor with the two types of operators

c) why is it important in a DNA footprinting assay to label only one strand of the DNA being tested?

Explanation / Answer

In lac operon contains three genes known as lac z that code for beta galactosidae ,lac y code for galactosidase permease and lac A that code for galactosidase transacetylase. The expression of genes in lac operon is controlled by operator and repressor. The expression of genes occur when inducer binds to repressor causing conformational change in repressor. The negative control occurs when repressor binds to operator.

In constitutive mutants induction process is absent, genes are active all the time without control over its expression.

The Oc operator binds to repressor with low affinity than the wild type repressor.
