Question

We perform microarray analysis to compare gene expression in a healthy person and a person with a genetic disease. RNA extracted from each person is reverse transcribed into cDNA using red fluorescent nucleotides for the healthy person and green fluorescent nucleotides for the person with the genetic disease. These cDNA samples are mixed and hybridized to the microarray. Below are results for eight genes from the microarray: Genes A and B – Green signal Gene C – Red signal Genes D and E – Orange signal Gene F – Light green signal Genes G and H – Yellow signal A) Which of the genes are expressed at higher, lower, or the same relative level in the person with the disease compared to the healthy person? Briefly explain your answer (2/3 credit). B) Suppose we discover that the causative mutation in the person with the genetic disease is an insertion of several bases that makes the promoter of a gene nonfunctional. Which of the genes in the list above, if any, could potentially have this mutation, based on the microarray results

Explanation / Answer

Genes A,B, AND F are relatively expressed more and the gene G,H are expressed at lower percentage .This is because the colour green during analysis reflects the gene that is responsible for the disorder .The genes that are showing intermediatory colors orange and yellow are the ones that show may sow mutation as they in the trasitory color phase between green and red.
