Question

Obesity is a serious health problem worldwide due in part to increased food intake and reduced physical activity. Obesity is associated with a variety of human diseases including type 2 diabetes and cardiovascular diseases. Selective and specific inhibitors of acetyl-CoA carboxylase have been proposed as potential anti-obesity drugs. a) What effect would an inhibitor of acetyl-CA carboxylase have on fatty acid synthesis and fatty acid oxidation? b) One such inhibitor of acetyl-CoA carboxylase is CABI. What structural feature of CABI makes it a potential acetyl-CoA carboxylase inhibitor?

Explanation / Answer

a) acetyl Co-A carboxylase converts Acetyl Co-A into Malonyl Co A and Malonyl Co A blocks the carnitin shuttle which is involve in the transportation of fatty acid into the mitochondria as a result oxidation of fatty acid will not take place and they will keep on accumulating and it will cause obesity. If the inhibitor of acetyl Co A carboxylase is given than it will inhibit the conversion of Acetyl Co A into Malonyl Co A and the carnitine shuttle will not be blocked and oxidation of fatty acids will take place and it will not led obesity to be happen.

b) biotin is responsible for the transfer carboxyl group to acetyl Co A which forms Malonyl Co A and CABI is a biotin analogue is is hydrophobic as a result it can cross membranes also.hence the CABI can bind instead of biotin to Acetyl Co A and will inhibit its activity. This structure is somewhat similar to the biotin structure.
