We isolate the human oncogene--->sequence it--->isolate genome of puffer fish--->annealing of puffer fish genome to separate the DNA strands--->hybridize with human DNA--->the homologous strand will hybridize with human DNA--->DNA sequence of complementary strand of puffer fish noted--->mRNA isolated from the fish with help of DNA--->cDNA constructed--->5' and 3' RACE[rapid amplification of cDNA ends] done---->they are used to find out full length cDNA of target gene without constructing a cDNA library.
