Question

For each of the E coli strains containing the lac operon alleles listed below, indicate whether the strain as I (inducible), C (constitutive) or U (unable to express beta-galactosidase and permease. (One letter should be inserted for each protein) beta-galactosidase Permease I^+O^+Z-Y^+/F'I^+O^c Z+Y+----------------------- I^+O^+Z+Y^+/F'I^-O^c Z+Y- ----------------------- I^+O^+Z-Y^+/F'I^-O^+ Z+Y- ----------------------- I^+P^-O^-Z^+Y^-/F'I^+P^+ O^c Z^-Y+----------------------- I^sO^+Z^+Y^+/F'I^-O^+Z^+Y^- --------------- You are looking at the expression of a novel protein in B cells called Gaga-1 and want to find out how over expression of the protein affects B cells in culture. You have the gene sequence and want to eventually do a Western Blot. However, you do not have an antibody to this protein, how will you detect if your plasmid, containing the Gaga-1 cDNA, effectively transfected into the cell, using a western blot?

Explanation / Answer

Biological role of the lac operon: It ensures that a cell dedicates resources to the production of enzymes involved in lactose metabolism only when lactose is available in the environment.

Constitutive expression: continuously expressed under normal cellular conditions

Inducible operons: Transcription is usually off and needs to be turned on

Negative inducible operon:

Control at operator = negative
Transcription is off
Normally off so needs to be turned on = inducible

Positive inducible operon:

Control at operon= positive=activator
Activator not bound to operator, transcription off
Inducer binds to inactive activator protein, alter shapes allowing it to bind transcription is on

The lac Z gene codes for beta galactosidase enzyme. The lac Y gene codes for permease. According to the question different types of mutations are given

a. ANS: In this condition lac Z gene is constitutive and lac Y gene is constitutive.

b. ANS: In this condition lac Z gene is constitutive and lac Y gene is inducible.

c. ANS: In this condition lac Z gene is inducible and lac Y gene is inducible.

d. ANS: In this condition lac Z gene is not expressed and lac Y gene is constitutive.

e. ANS: In this condition lac Z gene is not expressed and lac Y gene is not expressed.

Please note this I answed for first question.
