Question

A few solution/concentration/dilution questions. Please solve :)

1.You have a protein sample that has a concentration of 0.75 mg/ml. You need to load 10 micrograms of this sample onto a gel. How many microliters of sample do you need?

2. How do you prepare 1L of 20mM Tris pH 8, 1M NaCl from a 1M stock of Tris pH 8? (Hint: use dilution equation, no pHing required).

3. You have a 3 mg/ml protein sample. What is its concentration in micrograms/microliters?

4.You have a 5 mg/ml sample of GST (26 kDa). What is GST’s concentration in micromolar?

5. If you have 30 ml of protein at 4 mg/ml, and concentrate it to 5 ml, what is the protein’s final concentration?

6. Coca-Cola Classic has 40 g of high fructose corn syrup in a 12 ounce can (355 ml), which is 55% fructose and 45% glucose (both Mw = 180 g/mol). What is the molarity of glucose in a can of coke?

7. The human blood volume is 5 L. If someone has a blood alcohol content (BAC) of 0.10%, what is the molarity of ethanol (Mw = 46 g/mol, density = 0.789 g/cm3) in their body in millimolar?

8. How do you prepare a buffer containing all of the following: 450 ml of 75 mM Tris-Acetate pH 9.5, with 350 mM NaCl, 25% w/v MgCl2, and 4.5% v/v glycerol?

Explanation / Answer

There are multiple questions here. i am allowed to answer only 1 at a time. So I will answer question 1 for you. please ask rest as different question.
1.
concentration =0.75 mg/mL
mass needed = 10 microg = 0.01 mg
use:
volume = mass/ concentration
             = 0.01 mg / (0.74 mg/mL)
            =0.0135 mL
            =13.5 micro litre
Answer: 13.5 micro litre
