Question
Calculate the required volume of 0.25mg/mL BSA standard to reach to each concentration and fill the blanks in the table. Total volume in each tube is 250 microliters and each tube should have the same amount of PBS buffer as 5 microliters.
Table: Components for each protein standard solution of BCA assay
Please keep correct units.
Tube Number Concentration of BSA Standard (g/mL) Volume of 0.25mg/mL BSA standard (L) Volume DDI water (L) Volume of PBS Buffer (L) 1 Blank 0 245 5 2 10 5 3 20 5 4 30 5 5 40 5 6 50 5 7 80 5 8 100 5 9 150 5 10 200 5Explanation / Answer
We wil use formula,
C1V1 = C2V2
lets say we need a 250 microliter of 10 microliter/ml BSA concentration
original solution has 25mg/ml = 25000 microgram/ml concentration
25000 x V1 = 10 x 250
V1 = 0.1 microliter
Similarly we fill the other columns
Tube No. conc.. of BSA std.(microg/ml) Vol. of BSA std (microL)
1 0 0
2 10 0.1
3 20 0.2
4 30 0.3
5 40 0.4
6 50 0.5
7 80 0.8
8 100 1.0
9 150 1.5
10 200 2.0
