Question
Dilution Samples:
Need to make 200 uL of each dilution
We are doing serial dilutions:
For milk- 1:100, 1:1000, 1:2000, 1:4000 and 1:8000
For first supernatant- 1:100, 1:500, 1:1000, and 1:2000
For Whey- 1:100, 1:200, 1:400 and 1:800
For column fractions- 1:5, 1:10, 1:20, 1:40, and 1:80
And it has to be 200 uL total in volume.
I understand that for 1:100 you would use 2uL milk and 198 uL water. And then use 2uL of this and put it in 198 uL of volume for a 1:1000.
I do not understand how much to use from the 1:1000 to do 1:2000. ETC.
Explanation / Answer
Ans. 0.4 microlter form 1:1000 and dilute it by 199.6 microliter.
It can be easily understood as follows :
Since if you take 2 microliter from 1:1000 and dilute it by 198 it will be 1:10000.
Now if you take 1 microliter from 1:1000 and dilute it by 199 microliter it will be 1 :5000
Now if you take 0.5 microliter from 1:1000 and dilute it by 199.5 microliter it will be 1 :2500
Now if you take 0.4 microliter from 1:1000 and dilute it by 199.6 microliter it will be 1 :2000.
