Question

How much agarose would you need to weight to make 100 ml of a 1.2% agarose gel? How much 40X TAE buffer would you need to add to make 1 liter of 1X buffer? How much water would you need to add? I have 5M Tris buffer and need to make 500ml of 500mM Tris. How much 5M Tris do I need? One of the steps in genomic DNA isolation requires 70% ethanol to precipitate the DNA. How much 100% ethanol do I need to make 50ml of 70% ethanol? How much water do I need to add? I have a 50 mg/ml ampicillin stock and need to make 500ml of media containing 50ug/mL ampicillin. How many microliters of 50mg/ml ampicillin do I add?

Explanation / Answer

Ans-1- we will require 1.2 gms of agrose to make 100 ml of 1.2 percent agrose solution.

Ans-2- here v1c1 = v2c2

V1- 40x

C1- unknown

V2- 1 liter or 1000 ml

C2- 1x

So = (40) c2=( 1000) (1)

C2= 25 ml

So 25 ml of 40x buffer and than remaining water I.e. 975 ml

ans-3- one mole contain 1000 milimoles so

v1c1= v2c2

(5000) (c1)=( 500) (500)

V1= 50 ml.

Ans-4- v1c1= v2c2

(V1) (100)= (50) (70)

V1= 35 ml of 100% ethanol and 15 ml water.

Ans-5 one mg have 1000 ug so

V1c1= v2c2

V1 (50000)= (500) (50)

V1= 0.5 ml
