1. Ochterlony double diffusion:

i. Holes are cut in the agar, one central hole surrounded by other well

ii. Antibody is added to central well, antigens are added to the outer wells, the position of the band formed between the antigens allows for comparsion of the antigens to each other.

iii. It is useful for analysis of antigen and antibodies. An antigen specifically react with antibody to form antigen-antibody complex

iv.It is simplest techniques extensively used to identify out antisera for the presence of antibodies for a particular Ag and to determine titre.

v. These method is useful for detection and qualitative diagnostic procedures i.e., similarity between antigens,

Disadvantages:

i. It is semiquantative and long reaction time

2. Depending on similarity between antigens, different geometrical patterns are produced by antigen and antiserum wells. These pattern of lines can be interpreted to determine whether the antigens are same or different.

According to Ouchterlony plate image 2, show that:

i. 5 antigens and A is antiserum is added in the wells

ii. The antibodies in the antiserum react with both the antigens resulting in a smooth line of precipitate. Antigen 1, 2 and 3 are immunological identical. Where as antigen 4 and 5 are unrelated because their is no precipitation.
