Answer (2): to prepare lawn culture, spread plate and pour plate methods are used in which inoculum is spread using a spreader or poured along with media while plating thus, the growth obtained is spread like a lawn. whereas, to prepare a streak culture streaking is done in which a loop of wire (nichrome or platinum) is used to spread a culture inoculum in different streak patterns.

lawn culture is generally used to isolate a pure culture and to count colony forming units per mL of inoculum whereas, streaking is generally used to isolate a pure colony containing only one type of cells. both of these culture are routinely employed in microbiological laboratories fpr isolation of pure cultures and enumeration of number of organism presents by counting CFU/mL.

Answer (3) clear zone around antibiotics/disinfectants usually indiactes inhibition of growth of bacteria around the antibiotics. this clear halo around antibiotic discs is usually called as zone of inhibition because these chemicals inhibit growth of bacteria around them. these zones also indicates that the antibiotic/disinfectant used is effective against that bacteria and thus, used as antibiotic or disinfectant.

Answer (4) biofilms are mat of microrganism growing in layers stuck to each other by the extracellular matrix and polysaccharides produced by them during metabolic activties, biofilms are known to affect functioning of many instruments such as medical devces, water pipes etc and thus, affect human health. therefore, it is important to control biofilm formation. home humidifier is a good platform for microbes to form biofilms due to humid conditions.chemical disinfectants can be used to clean amd remove biofilms effectively.

Answer (5) During 1980s antibiotics were used widely in every house hold which created cross resistnce among microbes. antimicrobial resistance or popularly called as AMR increased during that time and still is occuring creating difficulties in treatment choices. antibiotic resistance developed due to over use of antibiotics by humans and thus resistance amonng bacteria, viruses developed due to genetic recombinations.
