The drug MTDIA is used to target the MTAP enzyme. A group treated with MTDIA showed an accumulation of MTA in both the A549 and H358 cancer cell lines. Why was there an accumulation of MTA when treated with MTDIA? I'm a bit confused with whether MTA is good or bad in cells. Why use a drug if MTA increases, when the purpose of the MTAP enzyme is to prevent the accumulation of MTA?
Also A549 and H358 treated with MTDIA showed a decrease in spermidine/spermine synthesis levels. Is this due to the increase in MTA inhibiting the product of polyamine biosynthesis?
Explanation / Answer
The drug is used in the treatment of human non-small cell lung carcinoma and human bronchioalveolar non-small cell lung carcinoma. The accumulation is because it is effective in treating both the above mentioned conditions. Also MTA levels in blood, urine increases due to MTDIA treatment.
