PCR amplify the region in which you expect to see an insert, looking for a larger band as compared to a vector control. Isolate the plasmid using a Qiagen kit and re-transform into another background strain. Analyze fluorescence under UV light. Isolate the plasmid using a Qiagen kit, and then digest with restriction enzymes to search for the appropriate sized insert.

For any gene construct we can screen it by doing restricition digestion, PCR and here in this cas we can analyze under UV light because the construct has GFP. Isolating the plasmid and re-transforming in to another background strain is not a screening method to check the clones. It is the next step in expression of the gene of interest.
