1)The DNA of a gene serves as a template for complementary base-pairing to form mRNA, catalyzes RNA polymerase II. The resulting mRNA is a single-stranded copy of the gene, which next then act as a template which is then translated at the ribosomal machinery.

2) There is two process of transcription termination in bacteria. Those are

Rho-independent termination also known as intrinsic termination in which RNA forms a hairpin structure which displaces RNA Polymerase and stops transcription and Rho-dependent termination that occurs when the Rho protein disassociates the RNA Polymerase and moves it off of the template.

3) The promotor binding site is different in the prokaryotic and eukaryotic system. That is why in order to express a eukaryotic protein in the prokaryotic system required promotor binding site specific to the prokaryotic system.
