Question

Calculate the final concentrations for both Chl a and Chl b.

Show data you used in your actual calculations (e.g. fresh weight of the tissue, volume of acetone added, 80% acetone used for dilution, OD645, OD663, etc).

Pay attention to the unit you will use to express your final chlorophyll a and chlorophyll b concentrations.

1) 80 % acetone

2) 1g = 1ml of fresh weight

3) dilution OD645, OD663,

Results

A= 0.11 T= 77 @ 645 nm

A= 0.19 T=65 @ 665

Calculate chorophyl a and chorophyl b

A645= (16.75ml/mg chla cm)[chla] (ln)+ (45.6 ml/mg chlb cm) [chlb] (ln)

A663 = (82.04ml/mg chla cm)[chla] (ln)+ (9.27 ml/mg chlb cm) [chlb] (ln)

Explanation / Answer

Hi
The formula to find out Chl concentration is:
Chlorophyll a (g/ml) = 12.7 (A663) - 2.69 (A645)
Chlorophyll b (g/ml) = 22.9 (A645) - 4.68 (A663)

Chl a = 12.7(0.19) - 2.69(0.11) = 2.413-0.2959 = 2.11 ug/ml
Chl b = 22.9 (0.11) - 4.68 (0.19) = 2.519 - 0.8892 = 1.62 ug/ml
