4 Add the following ingredients to each test tube Tabes 1, 4, and 7s 1 ml lipase, Im bile #Tubes 2, 5, and 8: 1 ml bold lipase, l ml bile Tubes 3, 6, and 2 ml water 5 Place tubes 1,2, and 3 in an ice-water bath, and incubase them for 30 minuces 6 Leave tabes 4,5, and 6 in your sest-tube rack to incuhate at room 7 Place tubes 7,8, and 9 in a warm-water bath set to 37 C, and incubate them for 30 minutes 8 After 30 minutes have passed, remove the subes from the ice-water bath and warm-water bath, and place them in temperature for 30 minutes your test-tube rack 9 Record the color of each tube in Table 2.4, and interpeet your resalts. TABLE 2.4 Enzymatic Activity of Lipase pH (Acidic, Neutral or Alaline 10 Answer the following questions about your results a What effect does temperature have on enzyme activity? b Were your fesults with the boiled lipase different from the results with nonboiled lipase? Why? Predict what color change you would see in tubes 3, 6, and 9 if you were to leave them in a warm-water bath for several days c 40 I Exploring Anatomy & Physiology in the Laboratory Core Concepts

Explanation / Answer

Temperature denatures enzymes as it breaks the tertiary structure of protein. This retards the enzymatic activity.

Boiled lipase will have different acitivity as compared to non-bioled lipase as boiled lipase will have reduced enzymatic activity.

Tube 3, 6, 9 will have no colour change as it does not have any lipase in it.
