Question
2. You are walking in the forest and discover a green "painted tongue" plant with no fungi attached. "Painted tongue" plants are usually red. However, a blue "painted tongue" plant has also been identified. Excited that you may have found a nevw fungicide you cancel your vacation and return to the lab. Self-fertilizations indicate your green plant and blue plants are homozygous, true breeding. Blue or green plants crossed to a wildtype red plant result in red plants. Since, the red and blues alleles of the "painted tongue" plant have not been shown to have fungicidal properties you suspect the green allele is a different gene. a) What experiment would you perform to prove this? Show your experiment b) What are your expected outcomes?Explanation / Answer
a) firstly i'll isolate the fungi and identify the morphological characteristic of DNA, then isolate the dna, perform restriction digestion with specific restriction enzyme, ligate it with specific enzyme, then run the pcr, then gene express to th both fungi.
Gene regulation can be done in two steps:
1- Identify the noble factor
2- Manipulate the transcription factor or manipulate the promoter region
b)outcome will be if there is functional gene (promoter + gene then protein-protein interaction will occur, then structure changes, morphology changes), then transcription occurs, and if there is no gene there will be no transcription.
