IV. Digital image analysis (DIA) is a common technique to quantify the DNA content in the cell nuclei on tissue and cells, which may help distinguish benign and malignant cells. As cell proliferation is one of the hallmarks in cancer cells, a higher DNA content can serve as an important indicator for cancer diagnosis. In DIA, cach nucleus of the cell on the slide are stained with an absorbing dye called Feulgen stain, which only binds to DNA, followed by the quantitative image analysis to measure the amount of DNA inside the cell nuclei. (1) Please propose the best type of microscope to use for this application. (2 points)Explanation / Answer
1- we can use mocrospectrophotometer, electron microscope, confocal fluorescent microscop, can use for use for this application.
2- quantitation of nucleic acid can be performed by the determination of concentration of DNA and RNA
There are two approaches which are use for determination these are.
Spectrophotometer quantitation and uv fluorescent tagging..
So the DNA content of 5 micron thick can determine by a formula.
A=abc.
By which we can get the dna content is 20 microlitter contain present in sample.
