2.) My question is : Can you make it simpler to understand this paper? State the general thesis, and the question the scientists are asking, and how did they approach the problem? Finally the conclusions they reached.
Explanation / Answer
At first let me point out the basic conclusions drawn
1) E89G mutation and M18V mutation are contrasting in nature.
2) The E89G mutation is enhancing where as M18V mutation has minimal effeect
3) M18V functions in dNTP binding and E89G functions as template primer grip.
Now this study is done to check how much the above mentioned mutations causes errors in RT-PCR and if one counter effects another?It was done on viruses that harbour this mutations.
I saw only one question in the discussion section
It said as this two mutations have very little effect on overall RT fidelity , does this mutations cause any great diffrenece when it comes to error specificity in those organism possesing this mutations?
The author answers , M18V it is very difficult to say. It is too little to assess. E89G has a 89G substitution , in vivo it was studied that virions with the enhancing effect in frameshift mutation and this 89G substitution ( they obviously will have an insertion or deletion) are more likely to be lethal than a point mutation.
