Question
Human Ph LAB REPORT 1. Prepare the following graphs showing the results of the lab exercises. Figures should be equipped with a title, axes labels (showing units if appropriate), and a legend if appropriate), and should be prepared using a computer a. Spectral Analysis of HBCN- Graph ABS vs wavelength (nm). Label lambda max on the graph. b. Standard Curve for HbCN- Show the regression line and formula for the standard curve. Indicate the ABS and the [Hb] for the unknown sample on the figure 2. Write an essay describing why the absorption spectra are different between oxyHb and deoxyHb. Discuss the problems this produces when attempting to measure blood Hb and the solutions to these problems. 3. Write an essay explaining how you would prepare from scratch a protocol to measure urine glucose by spectrophotometry 4. Data Sheet- Turn in a very neat copy of a completed data sheet. View as T |-5 -| /6Explanation / Answer
The oxyhemoglobin as well as deoxyglobin are two formsof hemoglobin. The oxy hemoglobin is the form she oxygen would be binding to the heme group of hemoglobin. Deoxygenated form is the hemoglobin form without oxygen. There is a difference in the absorption spectra of both these molecules. The oxyhemoglobin has lower absorption, approximately 660nm and de oxyhemoglobin has a wavelength of 940nm. This difference allows measurement of the oxygen amount in the blood of the patient. The instrument used to measure this is the pulse oximetry. This difference is due to the colours of the of the two molecules. Oxyhemoglobin is red in colourwhereas de oxyhemoglobin I’d blue in colour. This difference also explains for cyanosis, which is the bluish colour that develops as a result of hypoxia.
