Scenario 1:

a) The three controls which were missing from this experiment

1) JM109 (pM1); amp100

2) HY842; amp100

3) JM109 (pM1) + HY842; no abx

b) We cannot conclude that conjugation occurred between JM109 (pM1) and HY842 due to the missing controls JM109 (pM1); amp100 and HY842; amp100. These controls were highly important to understand which of these strains has ampicillin resistant marker. If we have these controls, we would have predicted the conjugation by their antibiotic resistance when grown in amp100 plates.

Scenario 2.

a) There is no conjugation happened between JM109 (pM1) and HY842 strains

Reason:

1) As we can observe from the first column that JM109 can grow in the presence of ampicillin (JM109 (pM1); amp100) but cannot grow in the presence of zeocin ( JM109 (pM1); Zeo100). This indicates that JM109 (pM1) has an ampicillin resistant gene but not Zeocin-resistant gene.

2) As we can observe from the second column that HY842 can grow in the presence of zeocin (HY842; Zeo100) but cannot grow in the presence of ampicillin ( HY842; amp100). This indicates that HY842 has a zeocin resistant gene but not an ampicillin-resistant gene.

3) As we can observe from the third column. After growing both JM109 (pM1) + HY842 together in the presence of Zeo100 and amp100 (JM109 (pM1) + HY842; zeo100+amp100), we cannot see any colonies. If conjugation has happened between both strains, a new strain would have formed with both amp and Zeo resistant genes which can grow in the presence of both amp and zeo.

As we did not observe any colonies in (JM109 (pM1) + HY842; zeo100+amp100) plates, we can conclude that no conjugation has occurred between these both strains
