1- Photophosphorylation is a process in which ATP is generated during the light reaction. During light reaction electrons and protons are generated by photolysis of water. The generated electrons are used by a cyclic electron transport chain to generate proton gradient. The generated proton gradient is responsible for the synthesis of ATP. Plastoquinone is the first electron acceptor in the light reaction from chlorophyll. If we block the plastoquinone activity then there will not any transfer of electron and ultimately no synthesis of ATP. When we add the electron acceptor E-grabber, the generated electron from photolysis of water, are grabbed by it and as a result of it, O2 is evolved.
2- During eclipse phase there is no light reaction. As we know that during the light reaction ATP and NADPH is generated which is utilized for CO2 fixation during the Calvin cycle. So During the short period of eclipse, there will not any generation of ATP and NADPH.
The first step of Calvin cycle is the fixation of atmospheric CO2 by RUBISCO enzyme. Ribulose 1,5-bisphosphate is broken down into Phosphoglyceric acid. This phosphoglyceric acid is then reduced with the help of NADPH into Glycerol-3 phosphate. Since there is NADPH and ATP the concentration of PGA will increase and the concentration of Ribulose 1,5-bisphosphate will decrease.
