Question
4. You are a researcher working on drug development and have synthesized a large number of molecules. After treating cultured human cells with the different drugs, you discover that one drug (Nowbeezinol) in particular changed the physiology of the cells in a way that could treat asthma. You decide to pursue this drug. When you chemically characterize Nowheezinol, you discover that it is very hydrophilic and is not capable of crossing membranes. A) Based on this, what can you conclude about the location of the receptor that Nowheezinol is interacting with? B) You study the biochemical interactions of Nowheezinol h cells more thoroughly, and find that the drug leads to activation of protein kinase A. What type of receptor does Nowheezinol appear to be interacting with? In comparison to the normal physiological state, what can you infer about the activation state of glycogen phosphorylase and glycogen synthase in the cells from this? C) You find that there is an antagonistic relationship beteeNowheezinol and a common heart medicine, Staynalivin. If Nowheezinol goes on the market and is administered to someone who is taking Staynalivin, what would you expect to happen? Would you recommend administering these two drugs together?Explanation / Answer
A) As Nowheezinol is incapable to cross the membrane its receptor must be located on the outside of the membrane.
B) Nowheezinol appears to be interacted with G-protein couple receptor as it activates protein kinase A.
If this drug intake it will activate protein kinase A which in turn activates glycogen phosphorylase and inactivates glycogen synthase. As a result glucose synthesis has increased from stored glycogen and it works antagonistic to insulin hormone.
C) As there is an antagonistic relation between Nowheezinol and heart medicine Staynalivin, Nowheezinol shows opposite action. That means Nowheezinol is not good for heart. Suppose heart medicine Staynalivin regulates heart, after taking Nowheezinol it disregulate heart.
It should never administer these two drugs together. Only heart medicine Staynalivin should administer.
