Question

I don’t have access to the appendix. The teacher never gave it to us. I need help answering the questions Note: BSA -bovine serum albumin, a blood protein of cattle A. Denaturation of proteins by heat, strong acids, or alcohol 1. Boil3 mL of a 2% BSA solution for a few minutes and describe the change in appearance. 2. Add 3 mL of water to11 mL of 2% BSA solu ion. Pour this solution into a tube containing 3 mL concentrated n layars. What happente at the Incerfacu betwsen the two layers? _d_adaent of95% ethanol to 3.mL of-24 SA·solution, shake well, and record any changes observed. Queations 1. For each "reagent" (EtoH, HNOs, and heat) a) name the type (a) of linkage which ia disrupted b) write one equation (use structure of side chains linked to a schematic protein backbone). Show "before and after" structures. (Xou may need to check your lecture text's section on denaturation for further information.) see p. 73- in Appendix for diagram of effect of ethanol hydrogen-bonding on protein denaturation. Do not copy 73E "verbatim". Use different AA R-groups.

Explanation / Answer

1.a. If we heat BSA it will denature in extreme temperature.BSA is a protein and the quarternary and tertiary structure is going to unfold to a primary linear structure by heat.Disrupting linkages are mainly hydrogen bonds and dispersion forces.

Nitric acid also break hydrogen bonds to make the protein loose it's active form.

Ethanol also does break hydrogen bonds of BSA and makes it separeted from the solution as precipitation.

b. The bond H-H is broken by heat in BSA .

HNO3 + H-H - Protein = H2O + NO2 - H - Protein .

EtOH + H - H - Protein = H2O + Et - H - Protein .
