Ans 1) The experiment was carried out by Lenski with the bacteria that were grown in a glucose limited minimal medium called as DM25. It was used as food source to obtain energy as it was used for isolating auxotrophic mutants of E.coli in presence of penicillin as selective agent. DM25 along with low concentration of glucose was supplemented and the DM24 consists of large amount of citrate. It was used initially to improve the killing efficiency during the experiments.
