Question

A very specific kind of early onset pancreatic cancer appears to be inherited in the Davis family. The disease took the late Mrs. Ruby Davis some years ago when she was 44 years old. Her son Jim, now 41, has just been diagnosed with this cancer. Learning this, his daughter Karen who is 18, went in for pancreatic tissue analysis, and there are early signs of DNA damage in some pancreatic cells.

Describe two gene therapy approaches (one in vivo involving viral delivery and one ex vivo involving T-cells) doctors might try to help Karen live a long and healthy life. You must say which genes are being targeted and provide the basics of how each approach works.

Explanation / Answer

In vivo involving viral delivery is a approach where genetic therapy is given inside the cells through vectors, the common vectors are virusus like cytomegalovirus, hepatitis in which the DNA of the virus is modified causing harmless and Gene modifications inserted into the virus making to replicate. It will make to correct the defects in genes of cancer cells.

Ex vivo involving T cell approaches outside of liver where tissue is obtained from liver and grown in culture. Adding specific genetic agents to correct the defects and make to grow. Once the new Gene repairing the defects is produced it is implanted in cancer cells make to grow send correct the remaining cells in liver.

Genes targeted in this therapy is Tumor suppression Gene. This Gene is responsible for cell growth, division etc. So loss of function of tumor suppression Gene leads to overgrowth of cells result in nroplasm.
