Explanation / Answer

To understand monoisotopic peak, first you have to understand what is monoisotopic mass,

So, Monoisotopic mass is the sum of masses of the atoms in a molecule using the gfround state , unbound , rest moass of the most abundant isotope for each element. For example , for carbon , the most abundant isotope is c-12 (which is lightest)

But some students got confused with this that always lighter isotop is considered which is totally wrong. LEet us understand with the help of a example that is IRON (Fe) , the most abu8ndant isotope of Fe is Iron-56 which is not the lightest , so relating it with the previous fact , you got understood that we have to consider the most abundant isotope.

So when you look for monoisotropic peak , look for the isotope with the smaller mass , that is , lighter element for smaller peptiudes. BUT if you consider larger peptides, the mono isotropic peak is not the most abundant peak in a spectrum, beecause the probability is higher for peptide to contain atleast 1 naturally occuring isotope.
