Question

What is the retention volume of a molecule with a molecular mass of 880,000 using the equation y = -15.75x + 117? _______ What is the molecular mass of a compound with a retention volume of 25 mL, using the equation y = -16.13x + 124? _______ For any chromatography technique where manually load the sample from the top of the column (i.e. with a pipet), why should you be careful putting in the solvent afterward? _______ When looking at the fractions collected in size exclusion chromatography, how can you tell where your sample is by just looking at it? _______

Explanation / Answer

4)

After the column is loaded, we have to open the stopcock and allow the solvent level to drop to the top of the packing, but do not allow the solvent layer to go below this point. Allowing this solvent level to go below the stationary phase, should always be avoided. Because it allows cracks, air bubbles and channel formation to occur leading to a poor separation.

The sample can be loaded directly to the top of the column. Normally, a minimum amount of a polar solvent, 5-10 drops, is used to dissolve the mixture.
