In drug discovery, affinity chromatography is used differently. Instead of ligating a ligand to the gel matrix and running a protein mix through the column, a target protein of medical importance is ligated to the gel matrix and a mixture of millions of different small organic molecules (called a chemical library) is run through the column. The purpose is to discover any compound that binds the target protein. The question for you is, after the column is charged and washed, what elution buffer can you use to retrieve the ligands from the column?
Saturated NaCl solution.
Concentrated HCl.
A membrane-binding protein.
Concentrated glucose slution.
Explanation / Answer
Saturated Nacl solution will be good elution buffer solution. This is so because the exchange process is highly pH dependent and buffer solution made out of Nacl doesn't change its pH value (as ionic strength remains same) with increasing concentration.
