1.) We will be separating cytosolic proteins during the DEAE column chromatography lab. (i) Why can’t we use the other subcellular fractions during this lab? (ii) What would we have to do to separate proteins from the other subcellular fractions?
3.) Three lab groups isolated subcellular fractions from liver tissue and three other groups
isolated subcellular fractions from brain tissue. The protein concentrations from the nuclear fraction of liver tissue measured 4.7, 6.3 and 5.9 mg/ml while the protein concentrations from the nuclear fraction of brain tissue measured 9.4, 10.5, and 8.9 mg/ml. (i) Are the values from the two different tissues statistically significantly different? (ii) Why or why not? Copy the table from your spreadsheet demonstrating your statistical calculations.
Explanation / Answer
1. (i) With DEAE chromatography, only negatively charged proteins and DNA molecules are separated.
(ii) For separating other proteins, we can use Isoelectric focussing i.e. separation on basis of pH.
