Lineweaver-burk plot need to be created for determination of KM and Vmax. The equation is given by

1/V =KM/Vmax)*1/S + 1/Vmax

so a plot of 1/V vs 1/S need to be created which gives a straight line. The slope is given by KM/Vmax and intercept is 1/Vmax.

In case of presence of an inhibitor ibuprofen, the equation still remains straight line but with slope of KMapp/Vmaxapp and intercept of 1/Vmaxapp.

V= rate of formation of PGG2. S= concentration of Arachidonic acid

So a plot of 1/V vs 1/S nee to be generated for both in the absence and presence of iuprofen.

In the presece of ibuprofen inhibitor, The equation of best fit is 1/V= (0.007)*1/S +0.023

1/Vmax app =0.023, Vmaxapp =1/0.023=43.5 mM/min and Slope = KM/Vmax= 0.007, KM=0.007*43.5 =0.3045 mM

since Vmax and Vmax app are same, the type of inhibition is competitive inhibition where binding of the inhibitor to the active site on the enzyme prevents binding of the substrate and vice- versa.
