Question

Calculate the amount of stock and buffer needed to make 100 uL of each target BGG diluted standard. You will need to know how to do this for an exam!!!!

To prepare 100µl of each dilution using 2.0 mg/ml (=2000ug/ml) BGG stock:

Target BGG Conc. (ug/ml)

2 mg/ml BGG stock (ul)

Homogenization buffer (ul)

0

200

400

600

800

40

60

1000

1200

1500

Be sure to use the same units for Cs (s=stock) and Cd (d=diluted) when using the equation”

Use CsVs = CdVd to calculate volumes above.

I'm missing something here and it's driving me nuts. I don't even know what BGG is and I feel like I need to know the molarity or molar mass of it. To try and work through it I tried to do the one that given and set it up like it says:

CsVs=CdVd   with Cs= stock concentration 2000ug/mL    Cd= target concentration 800 ug/mL and Vs=100ul

I end up with 250 for Vd. I don't know what I'm missing. I already have all the answers on the table but I have no idea how you are supposed to know that those numbers go there or where they come from.

Explanation / Answer

You do not need molar mass or molarity. Instead, convert the targeted BGG conc from ug/mL to ug/uL to make all the units same.

so, I am taking 800ug/mL as an example.

So the question says what volume of 2.0mg/ml BGG conc is needed to prepare targeted 800ug/mL.

so, convert ug/ml .

1ug/mL = 1 x 10^3 ug/uL

Now use CsVs = CdVd

Cs = 2x 10^6 ug/uL

Vs = ?

Cd = 800 x 10^3 ug/uL

Vs = 100 uL

so, Vs = 8E7/2E6 = 40uL

So, homogenised buffer needed = 100uL - 40 uL = 60uL

Now, you can calculated the rest varying Cd as the targeted BGG conc.
