Question
The enzyme invertase catalyzes the hydrolysis of sucrose to fructose and glucose. In a series of kinetic studies, you vary the concentration of the substrate, sucrose, and observe the following rates of product formation. Estimate the Km and Vmax values for invertase. Sketch a Michaelis-Menten plot consistent with your kinetic data. Label all salient features. A mixed inhibitor of invertase has recently been identified. Sketch a Lineweave Burke plot as a function of increasing inhibitor concentration. Label all salient features.Explanation / Answer
The Michaelis-Menten plot
y axis = velocity of reaction (V) in micro moles/min
x axis = substrate concentration, [S]0
Lineweaver-Burk plot is 1/V vs 1/[S]0
From the Lineweaver-Burk plot we have 1/Vmax = 0.5, therefore Vmax = 2.0
and -1/KM = - 0.025. therefore KM = 40
