Question
10. The drug Lipitor is an inhibitor of the enzyme HMG-CoA reductase and is therefore used in the treatment of high cholesterol. (20 Points) COOH F. Lipitor Briefly explain how you would interpret the following results? a. COOH HO HO NH same activity lipitor in vitro but inactive in vivo No Potency same pote ncy as lipitor Metabolism of Lipitor is known to primarily occur via P450 catalyzed oxidative hydroxylation of ring D. Provide a complete mechanism (using arrows) for this metabolic process and provide the structure for this metabolite of Lipitor. b.Explanation / Answer
Potent means its activity for a particular drug or you can say against a standard drug. Here structure 1 is showing same activity in vitro but inactive in vivo against the standard drug Lipitor. But structure 2 has no activity(because the ring A has been removed which shows its a pharmacophore for having the activity) where as structure 3 has same activity as standard drug.
