Question
In the lab exercise, you will load samples of anionic and cationic dyes onto a gel and than observe their migration through the gel when an electric field is applied, (a) W hen a sample o f DNA molecules is loaded onto a gel. will the molecules migrate toward the positive electrode or toward the negative electrode? When a sample containing a mixture o f proteins is loaded onto a gel. will the protein molecules migrate toward the positive electrode or toward the negative electrode? Although electrophoresis is a widely used technique for separating and analyzing mixtures o f nucleic acids or proteins, it is not very useful for analyzing most carbohydrates. Describe how a carbohydrate would behave in electrophoresis.Explanation / Answer
a)
Well ADN is a complex molecule and if you look for characteristic groups you will find that ADN has phosphate. These phosphates give a negative charge to the ADN, then when you place it in a gel and apply an electric field ADN will migrate to the positive electrode.
b)
Protetein could have negative or positive charge, then them could show a different migration depends of their charge.
c) Carbohydates don´t have any charge then elecphoresis is useless to perform a separation for these structures.
