Your research group was asked to investigate an enzyme, two substrates and a coenzyme given to you by a team conducting experiments to regulate their protein. Glutamate dehydrogenase (EC 4.2.6.3) is important not only in amino acid synthesis but also in the metabolism of alpha-ketoglutarate. The team conducted some experiments and found that the Km for substrate-1 went from 1.2x10^-2 units to 4.1x10^-3. This change was observed when guanosine triphosphate (a known effector) was added to the reaction mixture. An undergraduate student completed the experiment and sound that the Michaelis constant for substrate-2 went from 2.4x10^-2 units to 3.9x10^2 units after the effector was added. Also, the experiment showed that the maximum velocity did not change for both substrates.
