Question
The figure below is from an article examining the expression of miRNAs in glioblastoma multiform (GBM; Ciara et al., 2005). GBM tumors are highly invasive, aggressive and one of the most incurable of human cancers. How does the expression of each of the following miRNAs change in GBM tumours? (indicate whether each is under expressed, overexpressed. or unchanged) miRNA-221: miRNA-10b: miRNA-191a: miRNA-191b: miRNA-128: Make a hypothesis about how the change in miRNA-221 expression may lead to the development of cancer.Explanation / Answer
a. miRNA-221 is over expressed in all GBM tumors in comparision with control but it is under expressed in Hela cell lines and has no expression in neuroblastoma
miRNA-10b no significant differences in GBM & neuroblastoma in comparision with control but under expressed in Hela cells
miRNA-191a, miRNA-191b & miRNA-128 are downregulated in all GBM, Neuroblastoma and Hela cells in comparision with Control
b. Down regulation of miRNA-191a, miRNA-191b & miRNA-128 deregulates the cell cycle and promotes GBM
