Question

The lin-14 gene is a target of the lin-4 micro RNA. When lin-4 is expressed in C. elegans, lin-14 protein level! are dramatically reduced. You have specific inhibitors that block Drosha, Dicer and the RISC complex (a different inhibitor for each). If you treat late L1 stage worms (a developmental stage characterized by high levels of lin-4 expression) with each of the inhibitors, how would lin-14 protein levels be affected? Would your results be different if you also treated the worms with lin-4 pre-miRNA? In your answer, explain what each enzyme or complex normally does, why the inhibitor would have the effect that it does, and how the presence of lin-4 pre-miRNA would, or would not, change that.

Explanation / Answer

Answer-

Lin-14 gene expression will be decreased lin-4 micro RNA inhibits the expression of Lin-14 protein.

The result may be the same if you treat with Lin-4 pre-miRNA or precursor-miRNA, Beacuse from pre-miRNA, mi-RNA is formed after cleavage.

Usually, pre-miRNA is around 1000 nucleotide length, from which miRNA ( 17 - 24 nucleotides) get realeased by action of speciic enzymes. i.e Drosha.

Drosa is the double-strand-specific -ribonuclease which cleaves pre-miRNA at its hairpin like structure and then results the mi-RNA.

After the pre-miRNA transported to cytoplasm, it further cleaved by the enzyme Dicer.

RISC is the RNA induced silencing complex , by which miRNA is taken up and this protein-RNA complex act as factor to inerfere the RNA which is called RNA interference.

If inhibitors used for the either pre-miRNA or miRNA, of lin-4, then the expression level of Lin-14 gene/protein will be increased.
