Agar and gelatin are used as a solidifying agent for bacteriological media. Why is gelatin less suitable than the agar? 1. anaerobic bacteria are common cause of infection, some of which can be serious or life threatening. Because anaerobes are predominant components of the flora of the skin or mucous membranes, they are a common cause infections of endogenous origin. Because of their fastidious nature, anaerobes are hard to culture and isolate and are often not recovered from infected sites.
2. Blood agar is used because it is used to grow those organism which do not grow easily mostly fastidious bacterias. Other reason for using this media is it is used to grow wide range of culture.
3. Agar is preferred in media because agar stay solid at higher temperatures than gelatin and the other reason is that bacteria can digest gelatin more readily than agar.
4. presence of bubbles means the oxygen is released in the culture,it means isolated bacteria are able to produce catalase enzyme.so, they are strict aerobic or facultative anaerobic bateria. And the media on which oxygen bubbles are absent they are anaerobic bateria.
7. alpha hemolysis is also called as partial hemolysis. It is partial lysis of red cells in the media which produce green oxidized derivative methamoglobin while beta hemolysis called as complete hemolysis. It is complete lysis of red cell in the media around and under the colonies.
